Service Details

We offer a variety of single-molecule sequencing options on Nanopore's MinION or PromethION.

Plasmid-SIMPLE:

Our Plasmid-SIMPLE sequencing protocol includes transposon-based sequencing of an entire plasmid, no reference sequence required. This protocol requires 100-200ng of plasmid DNA as an input, and provides a minimum coverage of 100x. Customers will receive a fasta file with consensus, random positioning of the first base, and annotation from plAnnotate software. Original fastq files available upon request.

Plasmid-FS:

With higher sequencing depth and more detailed analysis, our Plasmid-FS protocol is perfect for challenging or unique plasmids, including long repeats and ITRs. Plasmid-FS orders include a unique cutter-based digestion and library preparation using Oxford-Nanopore's PCR-Free native barcoding kit. Sequencing coverage is 100x or up, as required by the client. 1-2 micrograms of plasmid input and a reference sequence are required for this protocol. Customers can expect to receive a consensus fasta file, a table of deviations from the reference, and a full basecall count with FRED quality sores. Original fastq files and BAM alignement are available upon request.
