Sequencing of difficult templates:

Occasionally, a submitted DNA template will not yield good quality data, when standard sequencing protocols are used. We refer to the template as "difficult." The following are the most common sequence combinations that render a template or a segment therein as difficult to sequence:
  • over 70% GC-rich regions
  • hairpins
  • long GC homopolymers
  • long poly A/T tails
  • di-and tri-nucleotide repeat stretches
  • long stretches of di-nucleotides that are not repeats.
  • long direct repeats

We have developed a number of proprietary protocols to effectively deal with each category of difficult templates. We ask that you inform us of difficult template, if you are aware of such conditions in your templates. It will speed up data turnaround and reduce your sequencing costs.
